With TP blood bank ELISA kit, the detection of TP antibodies is achieved by antigen "sandwich" enzyme-linked method (ELISA) where polystyrene microwell are pre-coated with recombinant Treponema pallidum antigens expressed in E. coli. The sample is added into microwells together with recombinant TP antigens conjugated to horseradish peroxidase (HRP-Conjugate). The pre-coated antigens express the same epitopes as the HRP-Conjugate antigens, but are expressed in different hosts. In case of presence of anti-TP in the sample, during incubation the pre-coated and conjugated antigens will be bound to the two variable domains of the antibody and the specific antigens-antibody immunocomplex is captured on the solid phase. After specific treatment with other reagents enclosed in these anti-Syphilis ELISA screening kits, the antigens-antibody immunocomplex will turn into yellow color, whose amount can be measured and is proportional to the amount of antibody in the sample. This indicate a positive result. Wells containing samples negative for TP antibody remain colorless.
