Anti-HIV 1/2 Two Step ELISA Screening kits use polystyrene microwell strips pre-coated with recombinant HIV antigens expressed in E.coli (recombinant HIV-1gp41, gp120 and recombinant HIV-2 gp-36). During the first incubation, the specific HIV1/2 antibodies, if any, will be captured. The microwells are washed to remove unbound serum proteins. A second recombinant antigen conjugated with Horseradish Peroxidase (HRP) is added. During the second incubation, this antigen will be bound to the captured antibody. The antigen-antibody-antigen (HRP) "sandwich" immunocomplex will turn into yellow with certain treatment with the reagents enclosed in the anti-HIV ELISA screening kits. The amount of color can be measured and is proportional to the amount of antibody in the sample. Wells containing samples negative for anti-HIV 1/2 remain colorless, which indicates a negative result. |
