HBV ELISA kits are performed on this principle (take HBsAg ELISA kit as example): HBsAg ELISA kit uses polystyrene microwell strips pre-coated with monoclonal antibodies specific to HBsAg. During incubation after serum or plasma sample is added along with a second antibody conjugated with horseradish peroxidase (HRP), an "Antibody-antigen-antibody sandwich immunocomplex" will form and be captured by the solid base, if the sample tested contains Hepatitis B Virus Surface Antigen, the HBsAg ELISA kit intends to test. After washing and other treatment, the well will turn yellow. The degree of the color can be measured and is proportional to the amount of antigen in the sample. Wells containing samples negative for HBsAg remain colorless.
